now we've collected our sample onto the grids the last stage is to add a final stain lead citrate and to add extra contrast to our sample i'm going to add a small amount of the lead citrate to a clean petri dish it's important not to breathe on the stain at this point because the carbon dioxide in our breath will cause stain deposits i'm just going to invert the sample on top of the drop of the stain there are sodium hydroxide pellets in the petri dish which will help absorb the co2 preventing stain deposits so now our samples rested for about three to four minutes we're just going to remove the grid from our drop of stain and wash off the excess in some filtered distilled water and this can then air dry before going for analysis on the electron microscope
